چکیده

During obesity, excess body weight is not only associated with an increased risk of type 2-diabetes, but also several other pathological processes, such as infertility. Adipose tissue the largest endocrine organ that produces adipokines, including adiponectin. Adiponectin has been reported to control fertility through hypothalamic–pituitary–gonadal axis, and folliculogenesis in ovaries. In this study, we focused on a recent adiponectin-like synthetic agonist called AdipoRon, its action human luteinized granulosa cells. We demonstrated AdipoRon activated adenosine monophosphate-activated protein kinase (AMPK) peroxisome proliferator-activated receptor alpha (PPAR) signalling pathways A 25 ?M stimulation reduced cell proliferation by inducing cycle arrest G1, PTEN p53 pathway activation. addition, perturbed metabolism decreasing mitochondrial activity ATP production. cells, phosphodiesterase activity, leading drop cyclic monophosphate (cAMP) production, aromatase expression oestrogens secretion. conclusion, impacted altering function, via steroid production proliferation. This may have applications for improving ovarian function metabolic disorders or cancers.

Regulation of Proliferation and Steroidogenesis in Human Granulosa Cells

Background and Aims:Human follicles selected for further development are thought to receive precise signals from gonadotropins and locally produced growth factors for survival. In this experiment, the mechanisms of the regulation of proliferation and steroidogenesis by GDF-9 and Br-cAMP in human ovarian cells were investigated. Methods:SVOG cells were cultured and treated with GDF-9 and Br-cAMP...

Effects of the insecticide amitraz, an alpha2-adrenergic receptor agonist, on human luteinized granulosa cells.

BACKGROUND Amitraz, an insecticide used to prevent tick and mite infestation of cattle, crops and dogs, is an alpha2-adrenergic receptor agonist that inhibits GnRH release and the ovulatory LH surge in rats. Noradrenalin, the physiological ligand for adrenergic receptors, inhibits progesterone production by IVF-derived granulosa cells, but the effects of amitraz are unknown. METHODS Luteinize...

Nitric oxide inhibits steroidogenesis in cultured porcine granulosa cells.

Recent evidence suggested that nitric oxide (NO) acts as an important factor in a variety of physiological and pathological roles, including reproductive functions. The purpose of the present study was to investigate whether NO might significantly induce any change in steroidogenesis in cultured porcine granulosa cells (PGC). An NO donor, NOC18, significantly suppressed the oestradiol release f...

Steroidogenesis in luteinized granulosa cell cultures varies with follicular priming regimen.

During follicular development, a co-ordinated gonadotrophin and endocrine environment is believed to be essential for normal function of the resulting corpus luteum. Whether differences in the gonadotrophins used to promote follicular development can have lasting effects on granulosa cells after they have undergone luteinization and culture, remains to be studied. We measured steroid production...

The effects of cetrorelix and triptorelin on the viability and steroidogenesis of cultured human granulosa luteinized cells.

BACKGROUND We investigated the effects of the gonadotropin-releasing hormone (GnRH) agonist triptorelin as well the GnRH antagonist cetrorelix those of on the viability and steroidogenesis in human granulosa luteinized (hGL) cell cultures. MATERIALS AND METHODS The hGL cells were obtained from 34 women undergoing ovarian stimulation for IVF treatment. The cells were cultured for 48 h with or ...
